Messenger RNA (mRNA)-based vaccines are advantageous because they can be relatively quicker and more cost efficient to manufacture compared to other traditional vaccine products. Lipid nanoparticles have three common purposes: delivery, self-adjuvanting properties, and mRNA protection. Faster vaccine development requires an efficient and fast assay to monitor mRNA purity and integrity. Microchip CE is known to be a robust technology that is capable of rapid separation. Here, we describe the development and optimization of a purity and integrity assay for mRNA-based vaccines encapsulated in lipid nanoparticles using commercial microchip-based separation. The analytical parameters of the optimized assay were assessed and the method is a stability indicating assay.  相似文献

We show that [6,6]-phenyl-C61-butyric acid methyl ester (PC61BM) at the surface of thin film blends of poly(3-hexylthiophene) (P3HT):PC61BM can be patterned by water. Using a series of heating and cooling steps, water droplets condense onto the blend film surface. This is possible due to the liquid-like, water swollen layer of poly(3,4-ethylenedioxythiophene) polystyrene sulfonate. Breath pattern water deformation and subsequent drying on the film surface results in isolated PC61BM structures, showing that migration of PC61BM takes place. This was confirmed by selective wavelength illumination to spatially map the photoluminescence from the P3HT and PC61BM. Within a device, redistribution of the surface PC61BM into aggregates would be catastrophic, as it would markedly alter device performance. We also postulate that repeated volume change of the poly(3,4-ethylenedioxythiophene) polystyrene sulfonate layer by water swelling may be, in part, responsible for the delamination failure mechanism in thin film solar cells devices. © 2015 Wiley Periodicals, Inc. This work explores coatings with thermally switchable wetting behavior, based on block copolymers that possess both hydrophilic and hydrophobic segments. The amphiphilic block copolymers were synthesized by coupling allyl-ended poly(ethylene oxide) (PEO) and hydride-ended poly(dimethylsiloxane) (PDMS) oligomers via a Pt catalyst. One near-symmetric diblock possessed an order-disorder transition temperature (TODT) of 64 °C. When cooled through TODT in ambient air, the PDMS domains wet the film's surface, producing a hydrophobic coating with a water contact angle (CA) = 90°. However, when cooled in humidified air, hydrophilic PEO domains form at the surface, yielding CA = 30–40°. The coatings can be reversibly switched between the two states by reheating above TODT, in the appropriate environment, and then cooling, rapidly generating the desired room-temperature surface wettability. © 2015 Wiley Periodicals, Inc. The charging and controlled oxidative doping of single organometallic ferrocene nanoparticles is reported in aqueous sodium tetrafluoroborate using the nano‐impacts method. It is shown that ferrocene nanoparticles of approximately 105 nm diameter are essentially quantitatively oxidatively doped with the uptake of one tetrafluoroborate anion per ferrocene molecule at suitably high overpotentials. By using lower potentials, it is possible to achieve low doping levels of single nanoparticles in a controlled manner.  相似文献
